No. From version 1.9.20 the back cover made for the ebook has no barcode box, because an ebook has no barcode. The space goes to the description and the author text. A back cover made with an earlier version is updated once, by itself, the next time you open the book. For the printed paperback, the cover wrap in the Covers page keeps KDP's exact barcode area clear, as KDP requires.
